Abstract:
The leaves of Khlu (Pluchea indica (Linn.) Less) were extracted and tested for
antioxidant activity using DPPH (1,1-diphenyl-2-picrylhydrazyl) scavenging assay.
It was found that the leaf ethanol extract exhibited a strong antioxidative activity with
an EC50 of 6.92 μg/ml.
The ethanolic extract of Pluchea indica leaf was further separated by Diaion
HP20 column to give four fractions, i.e the H2O fraction(EC50 = 16.02 μg/ml), the
H2O-MeOH fraction(EC50 = 2.65 μg/ml),the MeOH fraction (EC50 = 1.89 μg/ml) and
the EtOAc fraction (EC50 = 12.08 μg/ml). The most active fraction was selected for
further fractionation using silica chromatographic columns resulting in the isolation
of compound A1. Compound A1 was identified by spectroscopic techniques as
quercetin (5,7,3´,4´tetrahydroxyflavonol). Compound A1 is the major antioxidant
component in the leaf extract of P. indica with an EC50 of 1.69 μg/ml .

เมื่อสกัดส่วนใบของขลู่(Pluchea indica (Linn.) Less.) และนำไปศึกษาฤทธิ์
ต้านออกซิเดชั่นโดยวิธี DPPH scavenging assay พบว่าสารสกัดของใบมีฤทธิ์ต้านออกซิเดชั่น
ได้ดีโดยมีค่าEC50 = 6.92 ไมโครกรัม/มิลลิลิตร
เมื่อนำสารสกัดเอธานอลของใบขลู่ (Pluchea indica) มาแยกสารสำคัญโดยวิธี
Diaion HP20 คอลัมน์ ได้ fraction ต่าง ๆซึ่งแสดงฤทธิ์การต้านออกซิเดชั่นดังนี้ fraction น้ำ
(EC50 = 16.02 μg/ml), fraction น้ำ-เมทานอล(EC50 =2.65 μg/ml), fractionเมทานอล (EC50
=1.89 μg/ml)และfractionเอทิลอะซีเตท (EC50 = 12.08 μg/ml) นำ fraction เมทานอลซึ่ง
แสดงฤทธิ์ต้านออกซิเดชั่นที่ดีที่สุด มาแยกต่อด้วยซิลิกาเจลคอลัมน์โครมาโทกราฟฟี สามารถแยก
ได้สารA1ซึ่งพิสูจน์โครงสร้างโดยวิธีสเปคโตรสโคปีพบว่าเป็นquercetin(5,7,3´,4´
tetrahydroxyflavonol) สาร A1 เป็นสารสำคัญชนิดหนึ่งในสารสกัดเมธานอลที่แสดงฤทธิ์ต้าน
