Abstract:
The objective of this research was to determine the total phenolic content, antioxidant activities, chlorogenic acid and caffeine content, cytotoxicity and 5α-R inhibition activity of coffee berry extract and evaluate anti-hair loss efficacy of shampoo and tonic containing the extract on 54 volunteers. First the coffee berry was extracted with water, 25%, 50%, 75% and 95% ethanol. The best solvent was found in 75% ethanol with total phenolic content of 0.251 ± 0.002 mg GAE/ml extract, ferric reducing power activity of 0.296 ± 0.004 mg AAE/ml extract and DPPH radical scavenging with IC50 value of 0.546 ± 0.007 mg/ml. The extract showed high quantity of chlorogenic acid (34.820 ± 0.787 µg/ml) and caffeine (22.290 ± 2.732 µg/ml) by HPLC technique. It showed low toxicity to cells > 80% and 5α-R inhibition of 16.52% which was lower than finasteride and dutasteride (32.51% and 20.08%).
The stability of shampoo and tonic containing coffee berry extract (S6 and T6) was investigated by heating-cooling cycles (6 cycles) and constant temperature stability, including ambient temperature, 4ºC, 45ºC and under light exposure, for 12 weeks. At the end of the experiment, at 45ºC and the heating-cooling cycles affected a slight intense color of products. In addition, all storage conditions showed a slight change in pH value and viscosity for 12 weeks. Moreover, the products were classified as safe due to no microbial contamination and non-irritation on the skin.
In clinical trial, 54 subjects with ages 25 - 61 years and who experienced hair loss problems. The volunteers were randomly divided into placebo and CB groups. The CB group showed significant reduction in the mean number of hair loss after the combing test from 11.5 to 1.4 (p < 0.05) for 12 weeks. In hair photography, hair density in the CB group increased and new hair was observed under microscope analysis. The CB group showed 14.00% reduction of anagen hair loss by hair pulled test. In conclusion, the CB group showed better hair loss reduction efficacy than placebo. These results indicated that coffee berry extract can be used as anti-hair loss ingredient in hair care cosmetics
